Thidiazuran (TDZ) Induced Regeneration from Cotyledonary Node Explant of Abelmoschus moschatus Medik. L., (A Valuable Medicinal Plant)

چکیده

Knowledge concerning in vitro regeneration and developmental responses of Abelmoschus moschatus Medik. L. (muskdana plant) is limited. Aseptic seedlings were successfully raised on Murashige and Skoog (MS) basal medium augmented with GA (0.5 mg L ). The seeds did not germinate on MS medium alone. 3 1 Cotyledonary Node (CN) gave the best response with regard to induction of multiple shoots among the various explants used for morphogenesis. MS medium supplemented with 0.01 mg L Thidiazuran (TDZ) 1 proved to be the best for multiple shoot induction. A combination of 0.25 mg L N6 benzyladenine (BA) and 1 0.5 mg L Kinetin also gave better response but the number of shoots were comparatively less than those 1 induced on TDZ supplied medium. MS half strength supplemented with 0.5 mg L Indole-3-Butyric acid (IBA) 1 was found suitable medium for root induction in excised micro-shoots. The plants were successfully acclimatized in the green house at 70% survival rate.
